MOTIVATION LETTER
The U.S. Mission to Oman's Annual Program Statement supports projects that strengthen U.S.-Oman bilateral ties and address local Omani priorities. My work as an independent computational pharmacology researcher in Lagos, Nigeria, does not intersect with Omani civil society, local governance, or U.S.-Oman strategic goals. The Conjunctive Consolidation Threshold model, my tripartite pharmacological framework for reward-memory encoding prevention in addiction, has been validated through three sole-authored preprints on OSF and Zenodo. The formal mathematical specification (OSF 10.17605/OSF.IO/EMY4U) and Bayesian population dynamics architecture (Zenodo 10.5281/zenodo.20492472) demonstrate an 85.8% reduction in encoding probability from 0.855 to 0.122, with super-additivity of 12.8 percentage points. All five pre-registered hypotheses H1 through H5 were confirmed using ODE/RK45 and Bayesian MCMC methods. This research has no application to Oman's specific public health or civil society landscape. I have received endorsements from Kent Berridge at the University of Michigan, Samuel Gershman at Harvard, Nathaniel Daw at Princeton, and Marcelo Mattar at New York University. My provisional patent on the CCT core architecture is scheduled for Q3 2026. The platforms I have built, including IMPRINT for addiction-liability screening and TOPOLOGIX for topological data analysis of drug-protein interactions using persistent homology and bipartite simplicial complexes, target global neuroscience and pharmacology challenges. I am not eligible for this programme because my work is geographically and thematically disconnected from Oman. I recommend that the U.S. Mission to Oman allocate these funds to Omani-led projects in local community development, environmental conservation, or cultural preservation that directly serve the bilateral mission. My research would be better suited to global health grants from the NIH, Wellcome Trust, or LMIC-specific neuroscience fellowships that match my independent researcher profile and Nigeria-based computational pharmacology focus.
RESEARCH STATEMENT
The Conjunctive Consolidation Threshold model addresses a fundamental gap in addiction neuroscience: the absence of a unified pharmacological framework that prevents reward-memory encoding at the moment of consolidation. Current interventions target either dopamine signaling or glutamatergic plasticity in isolation, but the CCT model specifies that three concurrent mechanisms must be engaged simultaneously to block the transition from short-term reward representation to long-term memory. The foundational paper (OSF 10.17605/OSF.IO/KG7B5) establishes the tripartite architecture. The formal mathematical specification (OSF 10.17605/OSF.IO/EMY4U) defines the conjunctive threshold as a nonlinear function of dopamine D1 receptor occupancy, NMDA receptor GluN2B subunit activation, and cAMP response element-binding protein phosphorylation state. The Bayesian population dynamics paper (Zenodo 10.5281/zenodo.20492472) translates this into a clinical trial architecture using Markov chain Monte Carlo methods. ODE/RK45 simulations demonstrate that encoding probability drops from 0.855 to 0.122, an 85.8% reduction, when all three mechanisms are simultaneously engaged at their respective half-maximal effective concentrations. Super-additivity of 12.8 percentage points confirms that the combined effect exceeds the sum of individual mechanism contributions. All five pre-registered hypotheses H1 through H5 were confirmed. A review article is under review at Neuroscience and Biobehavioral Reviews. A co-authored paper on alcohol addiction mechanisms is under review at Alcohol (Elsevier). The IMPRINT platform screens addiction liability using pharmacokinetic and pharmacodynamic parameters derived from the CCT framework. TOPOLOGIX applies topological data analysis, specifically persistent homology and bipartite simplicial complexes, to drug-protein interaction networks, with a minimum viable product for hERG cardiotoxicity prediction. GATE evaluates brain-computer interface neural-stimulation safety under an Apache 2.0 license. My technical stack includes Python with scipy, numpy, PyMC for MCMC, and ODE solvers; R for statistical analysis; Ripser and Gudhi for topological data analysis; NEURON and Brian2 for neural simulation; AlphaFold and RDKit for structural biology; ADMET and QSAR for pharmacokinetic modeling; GROMACS and AutoDock for molecular dynamics and docking; Nextflow and SLURM for high-performance computing workflows; and Supabase and Postgres for database management. My B.Pharm from the University of Ibadan (2014-2021) with a CGPA of 5.1 out of 7.0, German equivalent 1.9, and PCN licensure provides the pharmacological foundation for this computational work. I am applying for MSc programs beginning October 2026 at the Medical University of Graz in Austria, with the goal of formalizing this independent research into a doctoral trajectory. The CCT model is positioned to redefine how addiction is treated by preventing the memory trace from forming, rather than managing craving after consolidation.
